Nanotechnologies for Intracellular Delivery of Cardiovascular Biotherapies

This talk will highlight recent developments in the Duvall lab on polymer-based nanotechnologies for intracellular delivery of biological drugs to improve cardiovascular bypass graft performance and promote tissue regeneration.

The first project that will be discussed focuses on a polymer-based “nanoplex” for efficient intracellular delivery of an anti-inflammatory and anti-fibrotic peptide drug to vascular transplant grafts. Our recent studies have shown that this nanomedicine decreases vasoconstriction and intimal hyperplasia in human saphenous vein and improves performance of vascular bypass grafts in a preclinical rabbit model.

The second part of the talk will highlight the development of a tissue engineering scaffold-based approach for local delivery of siRNA nanoparticles.  This system has been applied to effectively stimulate angiogenesis and tissue regeneration in vivo in subcutaneous implants and in diabetic rat excisional skin wounds.  These enabling technologies for intracellular delivery of biologics can potentially open up a new universe of targets previously considered to be “undruggable”, yielding safer and more effective therapies.
