Targeting Biotherapeutics in the Vascular System

Biotherapeutics including enzyme therapies gain increasing traction as a pharmacotherapy. They provide unique specificity and potency of action, yet their medical utility is restricted by rapid inactivation and necessity to be delivered precisely to the target sites in the body. We devise targeting approaches in which therapeutic enzymes are conjugated with affinity carriers providing anchoring and proper sub-cellular destination in the vascular endothelium and blood cells. This approach markedly improves the pharmacokinetics, targeting and effects of the enzyme therapies in animal models of oxidative stress, inflammation and thrombosis.
